These observations strongly support the notion that there is more than one suppressor on the long arm of chromosome 10 playing a role in uterine cancers. Careful evaluation of published LOH/deletion data suggests that there may be three tumor suppressor genes based on different regions and patterns of deletion in endometrioid tumors (115,117–119). A homeodomain gene, EMX2, was suggested as the candidate tumor suppressor mapping in the 10q25–q26 region (120). EMX2 mutations are Coukos01 001-034 11/18/04 16 1:32 PM Page 16 Goodfellow infrequent in endometrial cancers (10%) and additional studies are required to determine whether loss of EMX2 contributes to the development of uterine adenocarcinomas.
This syndrome was initially described by Aldred Warthin in 1895 when the striking family history of his seamstress, who died of endometrial cancer, came to his attention. In 1913, he published a paper describing this remarkable kindred, which included 10 cases of endometrial cancer and 7 cases of stomach cancer in 48 descendants of a man who died of cancer of the stomach or intestine (2). Multiple cases of colorectal cancer were noted in subsequent generations of this family (3). The HNPCC syndrome is also commonly called the Lynch Syndrome II, after Henry Lynch who systematically characterized this syndrome in the 1960s (4).
